When the first Guardians of the Galaxy movie was made, everyone involved knew that the audience was less familiar with Star Lord and the rest of the gang. They weren’t Captain America or Black Widow, after all.
So Chris Pratt suggested they make a joke of it, like no one in the movie knows who “Star Lord” is either. So in this scene, when he tells Korath he’s known as Star Lord, the latter says “Who?” – just like what most people in the theater were thinking at that moment.
